Аннотация: Callus photomorphogenesis processes of winter wheat species with different ploidy-level ( Т. monococcum L. (Au), Т. polonicum L. var. villosum (AuB) и Triticum aestivum L. (AuBD)) were analyzed. Calli had developed from mature and immature embryos on two types of media differed in auxin level. Photosynthetic activity (PA) of callus cultures was studied in dynamic in vitro on proliferation medium under optimal conditions as well, as under edaphic stress conditions (salinity - NaCl, 0,63% or drought - polyethyleneglycol 6000 16 % w/v). PA data fixed at photosynthetic photon flux density of 20 µmol·m-2·s-1 in «light curve» record mode was used for analysis of PSII effective quantum yield (Y(II)), quantum yield of non-regulated energy dissipated in PSII (Y(NO)) and quantum yield of non-photochemical quenching (Y(NPQ)) dynamics. Morphometric and cytological screenings of callus cultures were conducted. Species-specific of differences in callus color and tissue density was detected on induction medium. Calli developed from immature embryos showed PA as early, as they were passaged to proliferation medium. A numerous calli with chlorophyllcontaining areas (CCA) were detected under optimal conditions. Two picks in PA at 7th and 15th days were detected. The same picks were detected in our earlier work with soft spring wheat. Cytological analysis of callus tissue showed that stress conditions at proliferation stage result in plasmolysis, photomorphogenic tissue degeneration and CCA destruction primarily in calluses of less stress-tolerant species. In general, the most intensive process of photomorphogenesis under stress conditions was detected in calli which had characterized by small biomass buildup. The low levels of callus induction and PA as well, as high dispersion of PA data of calluses developed from mature embryos were described. So immature embryos are more suitable for the purpose of development of photosynthetic active callus culture of most part of species described in present article.
